چکیده

The nuclear factor-kB (NF-kB) pathway is important in neuronal survival and in integration of external signals e.g. cytokines, glutamate, Ab and nerve growth factor (NGF). During rat cerebellar development NF-kB activity is high in granule cells before postnatal day 7 (P7) and declines after P7. Using gene expression pro®les, measured by cDNA arrays, up-regulation of transforming growth factor-beta2 (TGF-b2) was correlated with the developmental down-regulation of NF-kB. TGF-b2 depicted strongest, more than 4-fold, up-regulation in P12 versus P4 cerebella. In situ hybridization and immunohistochemistry con®ned upregulated TGF-b2 to granule cells and correlated mRNA and TGFb2-protein increase. Finally TGF-b2 repressed NF-kB activity, in an in vitro system resembling migrating cerebellar granule cells. Thus, TGF-b might ful®ll an important role in repressing developmentally activated NF-kB in granule neurons. q 2001 Elsevier Science Ireland Ltd. All rights reserved.

TGF-β2 neutralization inhibits proliferation and activates apoptosis of cerebellar granule cell precurors in the developing cerebellum

Transforming growth factor beta 2 (TGF-beta2) plays a critical role in growth, differentiation and cell death, but its function in the developing cerebellum is still uncertain. In this study we analyzed the effects of TGF-beta2 on ex vivo developing cerebellar slice cultures. Proliferation of granule cell precursors peaked ex vivo in the same developmental window as in vivo (P8-P14). Addition o...

Trichothecin Induces Cell Death in NF-κB Constitutively Activated Human Cancer Cells via Inhibition of IKKβ Phosphorylation

Constitutive activation of the transcription factor nuclear factor-κB (NF-κB) is involved in tumorigenesis and chemo-resistance. As the key regulator of NF-κB, IKKβ is a major therapeutic target for various cancers. Trichothecin (TCN) is a metabolite isolated from an endophytic fungus of the herbal plant Maytenus hookeri Loes. In this study, we evaluated the anti-tumor activity of TCN and found...

TRANSFORMING GROWTH FACTOR β2 UP-REGULATES GM-CSF GENE IN HUMAN BLADDER CARCINOMA CELL LINE HTB 5637

Transforming growth factor betas are multifunctional polypeptides in the cytokine superfamily. They have a growth inhibitory role on hemopoietic progenitor cells in semisolid colony assay as well as in long-term bone-marrow culture. TGF - β2 represses stromal cells, stem cell factor gene transcription, and decreases the stability of c-kit transcripts in hemopoietic cells. TGF-β also modulat...

P75: Expression of GDNF Genes in the Cerebellum of Rat Neonate Born to Mother with Diabetes

Diabetes Mellitus as a common metabolic disorder in women of reproductive age is rising throughout the globe. Diabetes in pregnancy has various adverse outcomes on different organs development including the central nervous system (CNS) and it can cause learning deficits, behavioral problems and motor dysfunctions in the offspring. The cerebellum is a part of brain that coordinates voluntary mov...

Stereological Estimation of Granule Cell Number and Purkinje Cell Volume in the Cerebellum of Noise-Exposed Young Rat

In spite of the existing reports on behavioural and biochemical changes related to the cerebellum due to noise stress, not much is known about the effect of noise stress on the neuronal changes in the cerebellum. The present study aims at investigating the effects from one week noise exposure on granule cell number and Purkinje cell volume within the neonate rat cerebellum.15-day-old male Wista...
